| Abstract: | : | The rhythmic pumping of the mammalian heart arises from the intricately coordinated contraction of cardiomyocytes. However, the detailed three-dimensional organization of these cells has long been difficult to resolve. In this talk, I will introduce an integrated framework that applies computer vision methods to 3D confocal microscopy data to reconstruct cardiomyocyte orientation and geometry at micrometer resolution; an improvement of roughly three orders of magnitude over conventional approaches. I will show how this pipeline maps individual cardiomyocytes into coherent myofibers and further into higher-order laminar sheet structures that define ventricular architecture. By estimating orientation fields directly from volumetric microscopy data, we produce a comprehensive, multiscale reconstruction of myocardial organization at the cellular scale. Our work offers new insights into the structural basis of cardiac mechanics and establishes a foundation for high-resolution, data-driven models of heart function. |
